Learning Objectives: Determine optimal collection methods, and appropriate transport conditions for various bacterial and viral cultures (including fastidious and non-fastidious organisms). Summarize the advantages of a two bottle blood culture set and differentiate contaminant vs. pathogen with positive blood culture results. Utilize various stains (Gram, and Kinyoun) to determine bacterial morphology, establish specimen quality, and formulate differential diagnosis based on positive culture and stain results. Summarize the principles of susceptibility testing for bacterial pathogens. Explain several advantages as well as limitations of non-culture based detection methods, such as nucleic acid and antigen based methods for bacterial, viral, and parasitic pathogen detection. Review current best practice for diagnosis of HIV infection using both serologic and nucleic acid based methodologies.
